The aim of the project is to test the effect of topolin-type cytokinins (CK) as an alternative to N6-benzyladenine (BA) in the induction and proliferation of Norway spruce embryogenic cultures. BA and its degradation products are very stable. Among many other in vitro plant propagation systems, the negative effect of 9-glucoside is well known, inhibiting both growth and rooting of the resulting plants. CKs will be applied during the induction stage to overcome genotype-dependent recalcitrance, and during the proliferation stage to test whether they can increase the quantity and quality of somatic embryos produced and, subsequently, their germination.
